Towheed et al. 2005 Cochrane review (PMID 15846645) - 25 RCTs (4,963 patients) found glucosamine superior to placebo for OA pain (SMD -0.61) and function improvement. The crystalline glucosamine sulfate formulation (prescription-grade) showed stronger effects than over-the-counter glucosamine hydrochloride.$25-$45/month.

How to use

Take 1,500 mg/day of glucosamine sulfate once daily or in divided doses with food. Allow 8-12 weeks for assessment. Long-term use (3-6 months) is needed for maximum structural and symptom benefit.

How it works

Glucosamine sulfate is a precursor to glycosaminoglycans (the structural backbone of cartilage). Oral supplementation stimulates chondrocyte production of proteoglycans, inhibits cartilage-degrading matrix metalloproteinases, and has mild anti-inflammatory effects on synovial tissue.

Safety notes

Derived from shellfish - avoid if allergic to shellfish. May slightly raise blood glucose - people with diabetes should monitor carefully and inform their doctor.
